Advanced Plant Prime Editing Guide RNA Design Tool

Comprehensive tool for designing genomic pegRNAs with structure visualization for plant prime editing applications.

pegRNA design pipeline is currently limited to certain tools for accessing specific design components. PROpeg (Precision-optimized pegRNA) addresses these limitations by serving a unified platform implementing thermodynamic optimization, algorithmic scoring, linker identification, and secondary structure visualization functionalities. A standard pegRNA relies on a default scaffolding string. In contrast, our genomic prime editing guide RNA (gpegRNA) modifies the last three nucleotides of the 86-nt scaffold to perfectly complement the reverse transcribed region. This stable design prevents reverse transcription mismatches, minimizing unintended mutations and maximizing editing purity. The tool also facilitates gpegRNA engineered version (gepegRNA) design.
